Reactivity pattern is similar to previous reports for the “CMGWS supereplet” (Duquesnoy et al, Human Immunology 69: 826-832, 2008) and MICA group G1 (Zou et al, Immunogenetics 61: 91-100, 2009). In each of these earlier reports, limited panels were tested, and positive alleles shared residues 36C, 129M, 206G, 210W and 215S; Zou et al. suggested also that G1 includes 173K). The expanded panel tested by Mostecki showed reactivity directed to 36C; negative reactions were seen with 36Y-carrying MICA*005, *028, *042 and *051, which share one or more of residues129M, 173K, 206G, 210W and 215S.

This reactivity pattern was named MICA group 2 (MICA-G2) by Zou et al, defined by residues 36Y, 129V, 173E, 206S, 210R and 215T. To identify the specific epitopes required for a MICA-G2 antibody to react, hybrid recombinants of MICA*002 and MICA*008 were made by replacing residues at positions 173, 206, 210, 215 with residues of the other allele. The hybrid MICA*002 molecule carrying residues 173E/206S/210R/215T reacted with an eluate from MICA-G2-specific serum after absorption with MICA*009 transfected cells, while the hybrid MICA*008 molecule carrying residues of MICA*002 did not, showing that 36Y and 129V were not required for binding. No reactivity was seen with a hybrid recombinant MICA*002 that had 173E substituted for 173K, showing that this position also was not required for binding of that antibody.
